High parameter spectral flow cytometry: Guiding principles for successful panel design and experiment setup on the ID7000™ Spectral Cell Analyzer

With technology advances in high parameter flow cytometry, researchers are learning how to develop large (20+ parameters) flow cytometry panels for deep immune-profiling studies. The introduction of spectral cytometry analyzers has further accelerated scientists’ ability to use complex overlapping fluorochromes, giving them greater flexibility in panel design and allowing them to consider combinations that were previously challenging to resolve with conventional cytometers.

A good panel design is key to the success of a multicolor flow cytometry experiment, especially as the number of fluorochromes in a high parameter panel increases. Users must take into consideration the spectral properties and brightness of the fluorochromes, the relative expression levels of the antigens, marker co-expression, and commercially available fluorescent antibodies. The attributes of the instrument used for acquiring a high parameter panel also play a key role in the sensitivity and resolution of data.
